Cell reprogramming during liver regeneration

During embryonic development, animals generate many different types of cells, each with a distinct function and identity.

"Although the identities of these cells remain stable under normal conditions, some cells can be persuaded to take on new identities, through reprogramming," says Ben Stanger, MD, PhD, assistant professor of Medicine in the Division of Gastroenterology at the Perelman School of Medicine, University of Pennsylvania.

Researchers have been able to reprogram cells experimentally, but few have shown that cells can change their identities under normal physiological conditions in the body, particularly in mammals.

In the cover article of this month's issue of Genes and Development, Stanger, PhD candidate Kilangsungla Yanger, Yiwei Zong, PhD, and their colleagues, did just that in the liver of a mouse. Stanger is also an investigator in the Abramson Family Cancer Research Institute and the Department of Cell and Developmental Biology.

The adult liver contains two major cell types ? hepatocytes and biliary cells ? that differ dramatically in appearance and function. Hepatocytes are the main cell type in the liver, where they synthesize proteins and other macromolecules, and detoxify toxic substances. Biliary cells, on the other hand, line the bile ducts, which carry bile from the liver to the small intestine to help digest fats.

Using a sensitive method to tag and track how cells develop and differentiate, the researchers found that conditional expression of an activated Notch1 gene converted hepatocytes into biliary cells. Notch is an important receptor for relaying signals to tell cells how to develop.

What's more, after the researchers injured liver cells with a variety of toxins to stimulate wound healing, they found that over two to three weeks hepatocytes activated a biliary cell program on their own, acquiring the shape and function of biliary cells. These changes were dependent on the activation of endogenous Notch signaling.

"This is direct evidence that cells can be converted from one mature cell type to another in a live animal, as part of a normal response to injury," says Stanger. "We think that augmenting pre-existing cell reprogramming relationships may be another way to engineer cells for the treatment of diseases in which there are not enough bile ducts, such as cholestasis."

Yes, on some level it's just a seven-year check-in with people maybe half-remembered, if that. Yet the films also serve as a kind of check-in with us, too.

What ultimately is so compelling about 56 Up is the universality of the experiences. We were all once children. And we all will die. And in between, there is everything else.

We feel good, refreshed and depressed in watching these people get older, also embarrassed in moments and cautioned about the passage of time.

Apted, himself now in his early 70s, says he hopes to continue the series further. Long may it live.

Watching "56 Up" gives you the wonderful feeling of seeing a sociological experiment blossom into something novelistically rich and humane.

Time has been neither kind nor cruel to the 13 men and women profiled in "56 UP." It has just been time, which is what this groundbreaking series is about.

Chances are that you'll come away from this long film feeling a sense of knowing its characters.

We might say that '56 Up' serves much the same function as 'Amour,' but it responds to the inevitability of decline with compassion, not dread.

What started as a crafty way of looking at the U.K.'s rigid class structure has grown into a portrait of melancholy middle age, with its heartbreaks and minor-key triumphs.

Watching the eighth film is intriguing but, in a way, disappointing. At this point in the game, it feels as if all the characters have determined their lots in life and are simply plodding through their interviews.

Quite simply one of the great documentary projects in the history of cinema, an engrossing sociological experiment on film; and though this mostly mellow installment isn't as revelatory as some earlier ones, it's still a remarkable document.

... feels like a retrospective and summation of the whole series, with ample quotation from the previous films, an approach that makes it interesting even for viewers who haven't seen the previous installments.

A completely unique and remarkable documentary project.

Apted skillfully weaves old footage with the new, and we become poignantly aware of another factor shaping their lives (and our own): biology, as the we watch the once-cute kids grow gray and heavy.

Perhaps the boldest and probably longest running sociological experiment on film.

I think the best thing about this movie (and the entire series) is that it forces the viewer to think about their own lives. It's kind of an awakening experience.

Once again, Apted assembles a captivating documentary that's profoundly educational, essential viewing to aid the understanding of the human experience.

"56 Up" is well worth seeing.

56 Up is still moving and philosophic, though not as exciting as earlier episodes, which had more drama.

Syrian rebels capture key town near Jordan border

BEIRUT (AP) ? Syrian rebels on Friday captured a strategic town near the border with Jordan after a day of fierce clashes that killed at least 38 people, activists said, as opposition fighters expand their presence in the south, considered a gateway to Damascus.

The Britain-based Syrian Observatory for Human Rights said 16 rebels were among the dead in the fighting in and around Dael. The town lies less than 15 kilometers (10 miles) from the Jordanian border in Daraa province, where the uprising against President Bashar Assad's regime began two years ago.

The rebel gains have coincided with what regional officials and military experts say is a sharp increase in weapons shipments to opposition fighters by Arab governments in coordination with the U.S. in the hopes of readying a push into Assad's stronghold in the capital, Damascus.

Although rebels control wide areas in northern Syria that border Turkey, the Jordanian frontier is only about 100 kilometers (60 miles) from Damascus, or a third of the distance to the Turkish border.

The battle for Dael came as authorities ordered an investigation into a mortar attack on Damascus University that killed at least 10 students on Thursday, state media said. The attack was the deadliest since a wave of mortar shells began hitting the capital last month, puncturing the sense of normalcy the regime has tried to cultivate in the city.

It was unclear who fired the mortar rounds. The government blamed "terrorists," its blanket term for those fighting Assad's regime. Anti-regime activists accused the regime of staging the attack to turn civilians ? many of whom in Damascus are already wary of the opposition fighters ? against the rebels.

"Rebels now control wide areas in the Daraa countryside,'" said Rami Abdul-Rahman who heads the Observatory. "Every area that goes out of government control is important."

Syrian activist Maher Jamous, who is from Dael but currently lives in the United Arab Emirates, said that despite the steady advances and the latest rebel victory in Dael, the regime still maintains a strong presence in the strategic province that leads to the capital.

Jamous said the capture of Dael increases the pressure on the regime.

The regime is known to have posted elite troops in Daraa province, which separates Damascus from the Israeli-occupied Golan Heights that the Jewish state captured in 1967 and annexed in 1981.

Jamous said Dael has a population of 40,000, making it one of the bigger towns in the primarily agricultural region, which is dotted with small family farms. He added that the town fell briefly into the opposition's hands in the early days of the uprising, but was quickly retaken by regime forces in May 2011.

Amateur videos posted online by activists, showed rebels in the streets of Dael and the bodies of dead soldiers lying on the ground. The videos appeared genuine and corresponded to other AP reporting on the events depicted.

In other areas, the Observatory said heavy clashes were taking place between regime forces and fighters renewing their attempts to storm a strategic military facility, known as the 17th Division base, north of the city of Raqqa that was captured by rebels earlier this month.

The division is considered one of the most important remaining regime strongholds in the northern province that borders Turkey, the Observatory said. It added that warplanes carried out several air raids in the area.

The Observatory said regime forces bombarded the Damascus suburb of Adra, while the government al Al-Ikhbariya TV said troops killed "many terrorists" in the area which is close to one of the main jails in the country.

The Aleppo Media Center and the Observatory reported clashes, shelling and attacks by helicopter gunships near the international airport of the northern city of Aleppo, Syria's largest and commercial center.

Syria's crisis began in March 2011 with protests demanding Assad's ouster. Following a harsh government crackdown, the uprising steadily grew more violent until it became a full-fledged civil war. The U.N. says more than 70,000 people have been killed since.

Bungie shows off 'Destiny' characters on video

"Halo" creator Bungie has been sparing with details on the gameplay of its next big project, "Destiny," but liberal with concept art. The veil was lifted a little further Friday with the release of a video showing characters making the jump from concept to in-game model.

The video was shown at Bungie's panel at the Game Developers Conference in San Francisco. While it doesn't contain any actual gameplay, there's plenty of info to be extracted from the character models and animations.

For instance, the "Cabal" fighters are heavily armored and likely fill the role of tank. The three classes shown at the end ? Titan, Warlock, and Hunter ? fit neatly into the "warrior, wizard, rogue" archetypes already in place in thousands of games.

Sharp-eyed viewers will identify several influences, from "Warhammer 40K" and "Star Wars" to manga like "Appleseed" to, of course, Bungie's previous games. A batch of concept art was also released, which Kotaku has kindly collected here.

More substantial information about the game itself will likely be given out at E3 in June.

"Quit What You Suck At"When you suck at something, it's not a lot of fun but you've likely convinced yourself to keep going for one reason or another. We tend to get stuck out of habit, but sometimes you just need to quit. As venture capitalist Brad Feld often says, "quit what you suck at." He explains why in an interview with productivity and ideas blog the 99u:

If you are doing something poorly AND you don't enjoy it, then you probably suck at it. If you are doing it poorly, but it's important to you, or you want to get better at it, or it fascinates you ? keep trying. You usually know when you hate something ? that's a leading indicator that it's not worth doing, unless you have no other choice.

You are, however, going to suck at most everything the first time you try it and probably for a little while after that. This advice wasn't crafted to tell you to give up every time you suck, but rather to pay attention to what you hate and aren't any good at. When those two things coincide, and you've given that line of work/activity/whatever a decent change, you should quit or you'll continue making yourself unhappy.

(AP) ? Tiger Woods had gone more than three years without a World Golf Championship. He had no trouble remembering the drill.

He sat at a table with the blue Gene Sarazen Cup trophy ? he has six just like it at home ? and signed a stack of flags for posterity and charity after a two-shot win that was never in question during the final two hours on the Blue Monster at Doral.

Woods entertained a few questions about his new 5-wood, how his performance stacked up with a four-shot win in January at Torrey Pines and whether he thought Rory McIlroy, now overlooked as the No. 1 player in the world, was on the right track.

"Can the Masters get here soon enough?" someone else asked.

It was the only question Woods ignored.

The Masters is a month away, and Woods hasn't looked this equipped to win another green jacket in five years.

A year ago, he hobbled off the Blue Monster and was driven away in a cart after 11 holes of the final round because of tightness in his left Achilles tendon, raising questions about his health and his immediate future.

Sunday he left Doral on his own terms and with a record that is starting to look familiar.

Woods delivered two quick birdies to end the suspense, and two late bogeys only made the Cadillac Championship look closer than it was. Woods never let anyone get closer than three shots until it no longer mattered. Despite a conservative bogey on the final hole, he closed with a 1-under 71 for a two-shot win over Steve Stricker.

He now has won five times in the last year, the most of anyone in the world.

He goes to Bay Hill in two weeks for the Arnold Palmer Invitational, where he is the defending champion, with a chance to go to No. 1 in the world.

And he stamped himself as the favorite at the Masters.

"That's how I know I can play," Woods said. "That's the thing. To be able to bring it out a couple times so far this year ? and then be able to close and get the Ws on top of that ? that's nice. Any time I can win prior to Augusta, it always feels good."

Just like Torrey Pines, it was a helpless feeling for those trying to catch him.

Graeme McDowell, who also played with Woods in the final group at Bay Hill last year, again tried to apply pressure early in the round with a two-putt birdie and a beautiful approach into 7 feet on the second hole. Woods, who had to scramble for par on the easy opening hole, sank an 18-foot birdie putt on the second.

"Graeme hits it in there stiff, I need to answer," Woods said. "It was important to make that, and basically continue it."

And that he did.

Woods hit a high cut with an 8-iron into 4 feet on the par-3 fourth hole, and he was on his way.

Stricker chipped in for birdie on the 13th hole and kept trying to make up ground until he ran out of chances, and ultimately ran out of holes. He had a 68 to finish second.

"His attitude and what I saw this week, and his belief in himself again, looks very similar to when he was in the early 2000s, or you can pick any year when he was playing great," Stricker said. "He just seems to be in a better place mentally."

Woods was in a better place on the putting green, and he can thank Stricker for that.

They were going to play a practice round on Wednesday, but when Woods arrived at Doral, he couldn't find Stricker. Only after he finished playing and headed to the practice green did they meet.

Stricker offered a valuable tip for Woods, noticing that his posture was off when he stood over putts. They spent nearly an hour on the green that Wednesday afternoon, and Woods felt as good as he did at Torrey Pines. And it showed.

He took only 100 putts for the week, the fewest of his career on the PGA Tour. He made 27 birdies, one short of his personal best.

And he finished at 19-under 269 for the 76th win of his PGA Tour career. Not only is that six away from the record 82 won by Sam Snead, it's one more than Phil Mickelson and Vijay Singh combined.

"Thank you to Steve for the putting lesson," Woods said at the trophy presentation. "It was one of those weeks where I felt pretty good about how I was playing, made a few putts and got it rolling."

Stricker, playing a part-time schedule, picked up his second runner-up finish in just three starts. He had no regrets about offering Woods some help.

"At times you kick yourself," Stricker said with a laugh. "He's a good friend. We talk a lot about putting. It's good to see him playing well."

Asked if he would have won without that chance meeting with Stricker, Woods hedged.

"I would like to say I probably would have, but ..." he said with a smile. "I've been putting at home and it just still hadn't felt right. I still was a little bit off. ... He basically got me in the same position that I was at Torrey. So once he put me in there where I felt comfortable, I said, 'Well, this is not too foreign. This is what I was a month or so ago.' And I started rolling it and it felt really, really good."

McDowell had third place to himself until he went for the green on the 18th hole and found the water. He made double bogey, shot 72 and fell into a four-way tie for third that cost him $172,500. Phil Mickelson (71), Sergio Garcia (69) and Adam Scott (64) also tied for third.

Woods improved to 41-2 on the PGA Tour when he had the outright lead going into the final round, the last two wins with McDowell at his side. Woods last won while ahead at Bay Hill a year ago.

"The way Tiger was playing, I was always in chase mode," McDowell said. "He was always going to be a tough guy to catch. Fair play to him. He played fantastic golf the last couple of days."

McIlroy's week ended on a happy note.

Not only did he finish the tournament, he might have turned the corner with a bogey-free 65. McIlroy opened with a 7-iron into 18 feet for eagle, which he called one of the best shots he hit. He shot a 32 on the back nine for a round that surprised him considering how far away he felt when he arrived at Doral.

"Just goes to show, it's not as far away as you think," McIlroy said. "That's been one of my problems. I always think when I'm playing bad that it's further away than it is. That's just where I have to stay patient ... and know that if I put in the hard work, that the results will bear fruit. Whether that's sooner or later, it doesn't really matter."

The Masters is approaching quickly, and Woods has reason to be excited about it. For now, he cared only about his latest trophy.

"Majors and World Golf Championships are the best because you know you are playing against the best players," Woods said. "That's what makes wins like this special. That's why I love to compete."

S. Korea, U.S. begin drills amid North's threats

SEOUL, South Korea (AP) ? South Korea and the United States began annual military drills Monday despite North Korean threats to respond by voiding the armistice that ended the Korean War and launching a nuclear attack on the U.S.

After the start of the drills, South Korean officials said their northern counterparts didn't answer two calls on a hotline between the sides, apparently following through on an earlier vow to cut the communication channel because of the drills.

Pyongyang has launched a bombast-filled propaganda campaign against the drills, which involve 10,000 South Korean and about 3,000 American troops, and last week's U.N. vote to impose new sanctions over the North's Feb. 12 nuclear test. Analysts believe that much of that campaign is meant to shore up loyalty among citizens and the military for North Korea's young leader, Kim Jong Un.

Pyongyang isn't believed to be able to build a warhead small enough to mount on a long-range missile, and the North's military has repeatedly vowed in the past to scrap the 1953 armistice. North Korea wants a formal peace treaty, security guarantees and other concessions, as well as the removal of 28,500 U.S. troops stationed in South Korea.

Still, South Korean and U.S. officials have been closely monitoring Pyongyang's actions and parsing the torrent of recent rhetoric from the North, which has been more warlike than usual.

North Korea regularly claims South Korea-U.S. drills are a preparation for invasion, but Pyongyang has signaled more worry about the drills that began Monday. The drills follow U.N. sanctions that the North says are the result of U.S. hostility aimed at toppling its political system.

North Korea has also warned South Korea of a nuclear war on the divided peninsula and said it was cancelling nonaggression pacts.

Under newly inaugurated President Park Geun-hye, South Korea's Defense Ministry, which often brushes off North Korean threats, has looked to send a message of strength in response to the latest threats. The ministry warned Friday that the North's government would "evaporate from the face of the Earth" if it ever used a nuclear weapon. The White House also said the U.S. is fully capable of defending itself against a North Korean ballistic attack.

North Korea has said the U.S. mainland is within the range of its long-range missiles, and an army general told a Pyongyang rally last week that the military is ready to fire a long-range nuclear-armed missile to turn Washington into a "sea of fire."

While outside scientists are still trying to determine specifics, the North's rocket test in December and third atomic bomb test last month may have pushed the country a step closer to acquiring the ability to hit the U.S. with weapons of mass destruction. Analysts, however, say Pyongyang is still years away from acquiring the smaller, lighter nuclear warheads needed for a credible nuclear missile program.

But there are still worries about a smaller conflict. North Korea has a variety of missiles and other weapons capable of striking South Korea. In 2010, North Korea shelled a South Korean island and allegedly torpedoed a South Korean warship, killing a total of 50 South Koreans.

Both incidents occurred near the disputed western sea boundary, a recurring flashpoint between the Koreas that has seen three other bloody naval skirmishes since 1999.

Kim Jong Un visited two islands just north of the sea boundary last week and ordered troops there to open fire immediately if a single enemy shell is fired on North Korean waters. Kim was also quoted as saying his military is fully ready to fight an "all-out war" and that he will order a "just, great advance for national unification" if the enemy makes even a slight provocation, according to the North's official Korean Central News Agency.

Despite the threats, South Korea and the U.S. began the 11-day war games as scheduled Monday. The allies have repeatedly said the drills and other joint exercises are defensive in nature and they have no intention of attacking the North.

A U.S. military statement said the exercise is not related to current events on the Korean Peninsula.

The drills are part of larger war games that began March 1 and are set to go on for two months.

U.S. troops in South Korea are meant to prevent North Korean aggression, U.S. and South Korean officials say, and are a legacy of the Korean War, which ended with the peace treaty that leaves the Korean Peninsula still technically in a state of war.

The former vice president got straight to the point in R.J. Cutler's latest documentary "The World According to Dick Cheney," telling the filmmaker he doesn't care what his critics think.

"I don't lay awake at night thinking 'gee, what are they going to say about me now?'" Cheney remarks in the upcoming film.

In an interview forABC's "This Week," Cutler responded "He does say a lot that he's not interested in what people think about him, but it's hard to imagine that he's not invested in what his legacy is. He is a significant figure of American history."

The documentary, which premieres March 15 on Showtime, features an extensive interview with the retired politician and offers a rare glimpse into Cheney's life since leaving Washington.

Cutler said he was strategic in approaching Cheney about appearing on the big screen.

"I was advised early on that the best path to getting him to participate would be patience," Cutler said. "And indeed it took seven months between the time that I first reached out to him and the time that he invited me to have lunch with him to discuss what my plans were for the film."

The director, whose previous documentaries include "The War Room" and "A Perfect Candidate," said he was driven by a desire to find out more about the polarizing political figure.

"Making a film like 'The World According to Dick Cheney,' you need to enter most of all with curiosity," Cutler said. "Not with expectations, not with preconceived notions, but with questions."

After many lengthy interviews with Cheney, and even accompanying him on a fishing excursion, Cutler gained unique insight into the former vice president's political strategy.

"He does not feel there is room for compromise," Cutler said. "I think it raises the question, when total conviction serves a democracy and when it can be problematic for democracy. And that's a question that, to me, is worth considering not only in the specific analysis of the George W. Bush presidency and his relationship with Vice President Cheney and Vice President Cheney's career, but in thinking about democracy from a larger view. And so, this was a major reason why we wanted to make this film and something that I was really excited about exploring.."

Comet Pan-STARRS is closest to Sun today: See it at sunset

A?comet sailing through the inner solar system make its closest approach to the sun and will be at its brightest at sunset tonight, but the glare of twilight may make it tricky to see, NASA says.

The?Comet Pan-STARRS?will be 28 million miles (45 million kilometers) from the surface of the sun when it swings around the star today, and should be bright enough to see without the aid of telescopes or binoculars, weather permitting. But the comet is also appearing low on the western horizon at sunset so some planning is needed to spot the celestial wanderer with the naked eye tonight.

"Look too early and the sky will be too bright," said Rachel Stevenson, a NASA Postdoctoral Fellow at the agency's Jet Propulsion Laboratory in Pasadena, Calif. "Look too late, the comet will be too low and obstructed by the horizon. This comet has a relatively small window."

A good time to look is about 40 minutes after sunset. The comet may appear as a sort of exclamation point in the evening sky, with the point being the comet itself and its diffuse tail stretching nearly straight up from the horizon, JPL officials added. [How to see the comet]

Comet Pan-STARRS, officially known as comet C/2011 L4 Pan-STARRS, was discovered in June 2011 by astronomers using the Panoramic Survey Telescope and Rapid Response System (or PAN-STARRS), a telescope atop the Haleakala volcano in Hawaii. The comet takes more than 100 million years to orbit the sun and appears to come from the Oort cloud, a vast halo of comets and icy objects at the outer edge of the solar system.

While the comet is making its closest pass by the sun tonight, the best views of the object are still to come. NASA scientists said the comet's proximity to the sun may make it too difficult to spot tonight, but that will change over the next few days.

"As it continues its nightly trek across the sky, the comet may get lost in the sun's glare but should return and be visible to the naked eye by March 12," JPL officials explained. "As time marches on in the month of March, the comet will begin to fade away slowly, becoming difficult to view (even with binoculars or?small telescopes) by month's end."

And there is another reason to look for comet Pan-STARRS later this week. On Tuesday (March 12), the moon rises into the cosmic display.

"The comet will be joined in the western sky after dark by the slender crescent moon on March 12, 13, and 14," the editors of StarDate Magazine, a stargazing publication of the McDonald Observatory at the University of Texas. "Good naked-eye views of the comet should continue for several nights, with the comet remaining visible through binoculars into April."

Comet Pan-STARRS is one of three?comets?capturing the attention of stargazers this year. The Comet Lemmon C/2012 F6 is currently visible to observers in the Southern Hemisphere and at times was in the night sky at the same time, allowing stargazers to capture?rare photos of two comets in the sky together.

Meanwhile, the Comet ISON is making its way into the inner solar system and could put on a spectacular cosmic display later this year. Officially designated C/2012 S1 (ISON), Comet ISON was discovered in September 2012 by Russian amateur astronomers Vitali Nevski and Artyom Novichonok using a remotely operated International Scientific Optical Network (ISON) telescope.

Some astronomers have billed?Comet ISON?as a potential "comet of the century" since it could be brighter than the full moon in daylight when it makes its closest approach to the sun on Nov. 28. At that time, Comet ISON will be much closer to the sun than Comet Pan-STARRS is now. ISON will approach within 800,000 miles (1.2 million km) of the star, making it a true sungrazing comet.

The answer is, for now, no.? Once the Ravens realize that Boldin is serious about playing elsewhere, the answer could change to yes.

Afghan bombers strike during US official's visit

KABUL, Afghanistan (AP) ? Militants staged two suicide attacks that killed at least 19 people on Saturday, the first full day of U.S. Defense Secretary Chuck Hagel's visit to Afghanistan. They were a fresh reminder of the challenges posed by insurgents to the U.S.-led NATO force as it hands over the country's security to the Afghans.

"This attack was a message to him," Taliban spokesman Zabiullah Mujahid said of Hagel, in an email to reporters about one of the bombings, outside the country's Defense Ministry in Kabul. He did not comment on the other attack.

Hagel was nowhere near the Kabul blast, but heard it across the city. He told reporters traveling with him that he wasn't sure what it was when he heard the explosion.

"We're in a war zone. I've been in war, so shouldn't be surprised when a bomb goes off or there's an explosion," said Hagel, a Vietnam War veteran. Asked what his message to the Taliban would be, he said that the U.S. was going to continue to work with its allies to insure that the Afghan people have the ability to develop their own country and democracy.

In the first attack, a suicide bomber on a bicycle struck outside the Afghan Defense Ministry early Saturday morning, just as employees were arriving for work. About a half hour later, another suicide bomber hit a joint NATO and Afghan patrol near a police checkpoint in Khost city, the capital of Khost province in eastern Afghanistan, said provincial spokesman Baryalai Wakman.

Nine Afghan civilians were killed in the bombing at the ministry and 14 wounded, and two Afghan policemen and eight children died in the blast in Khost while another two Afghan civilians were wounded, according to a statement from President Hamid Karzai's office.

Karzai condemned the bombings, calling them un-Islamic. "The perpetrators of such attacks are cowards who are killing innocent children at the orders of foreigners," he said in a statement emailed to reporters. Karzai usually uses the term "foreigners" to refer to Pakistan, which he blames for failing to crack down on Taliban militants who take sanctuary there.

Hagel's first visit to Kabul as Pentagon chief comes as the U.S. and Afghanistan grapple with a number of disputes, from the aborted handover of a main detention facility ? canceled at the last moment late Friday as a deal for the transfer broke down ? to Afghan President Hamid Karzai's demand that U.S. special operations forces withdraw from Wardak province just outside Kabul over allegations of abuse.

Karzai released a statement late Saturday saying the transfer of the prison to Afghan control would go ahead in the coming week. It was unclear what derailed Saturday's ceremony. U.S. officials declined to comment on the statement, which was released after he met with top military commander in Afghanistan Gen. Joseph Dunford this evening in Kabul

The prison transfer, originally slated for 2009, has been repeatedly delayed because of disputes between the U.S. and Afghan governments about whether all detainees should have the right to a trial and who will have the ultimate authority over the release of prisoners the U.S. considers a threat.

The Afghan government has maintained that it needs full control over which prisoners are released as a matter of national sovereignty. The issue has threatened to undermine ongoing negotiations for a bilateral security agreement that would govern the presence of U.S. forces in Afghanistan after the current combat mission ends in 2014.

Regarding Wardak, Karzai set a deadline for Monday for the pullout of the U.S. commandos, over allegations that joint U.S. and Afghan patrols engaged in a pattern of torture, kidnappings and summary executions.

"Each of those accusations has been answered and we're not involved," said Brigadier Adam Findlay, NATO's deputy chief of staff of operations, in an interview with The Associated Press Saturday. "There are obviously atrocities occurring there, but it's not linked to us, and the kind of atrocities we are seeing, fingers cut off, other mutilations to bodies, is just not the way we work."

Findlay said NATO officials have made provisional plans to withdraw special operations forces, if Karzai sticks to his edict after meetings this weekend with Hagel and top military commander in Afghanistan Gen. Joseph Dunford.

"What we've got to try to do is go to a middle ground that meets the president's frustration," but also keeps insurgents from using Wardak as a staging ground to launch attacks on the capital, Findlay said. "That plan would be that you would put in your more conventional forces into Wardak," to replace the special operators and maintain security, he said.

NATO officials see the weekend violence as part of the Taliban's coming campaign for the spring fighting season. "There's a series of attacks that have started as the snow is thawing. We had a potential insider attack yesterday ... and there's been a number of attacks on the border," Findlay explained.

The suspected insider attack occurred in Kapisa province in eastern Afghanistan several hours before Hagel arrived Friday. Three men presumed to be Afghan soldiers forced their way onto a U.S. base and opened fire, killing one U.S. civilian contractor and wounding four U.S. soldiers, according to a senior U.S. military official.

The official said investigators were "95 percent certain it was an insider attack," because the three men came from the Afghan side of the joint U.S.-Afghan base, and rammed an Afghan army Humvee through a checkpoint dividing the base, before jumping out and opening fire on the Americans with automatic weapons. All three attackers were killed.

The official spoke on condition of anonymity because he was not authorized to speak publicly.

The Taliban said it was not behind the Tagab base attack, and has not yet weighed in on the attack in Khost, but the group claimed responsibility for the morning attack at the ministry shortly after it happened.

New England residents hunkered down Friday as a late-season storm dumped up to a foot of snow in some areas ? with more to come.

The storm also brought high winds that were battering Nantucket, Martha?s Vineyard, and Long Island, the Weather Channel reported.

A winter storm warning remained in effect for the region through 1 p.m., with snowfall expected to lessen through the afternoon.

?We are watching a conveyor belt of wave after wave of snow coming in over the Atlantic,? Alan Dunham, meteorologist with the National Weather Service, told the Associated Press. ?The morning commute will definitely be a challenge.?

The Massachusetts town of Mansfield reported 14 inches on Friday and Rockland had a foot, according to the National Weather Service. A seaside house on Plum Island, about 40 miles north of Boston, was listing at a 45-degree angle after being battered by waves, WHDH reported.

The accumulation of wet snow on trees and power lines combined with gusty winds could lead to some isolated power outages, NBC Connecticut cautioned. Hundreds of schools closed or delayed openings.

The weather meant a change of footwear for Lisa Parisella of Beverly, Mass., where there were six inches on the ground on Friday morning. ?I was thinking, March, ready to take out the sandals, and I?m taking out the boots again,? she said.

The storm has authorities along the Massachusetts coast worried as wind-lashed seas threatened flood-prone areas still recovering from last month?s blizzard. The town of Scituate, Mass. advised residents to leave low-lying zones during high tides on Thursday.

?I think that?s going to be very dangerous,? Scituate Police Chief Brian Stewart told the AP. ?We?re recommending that peoples in areas that have experienced coastal flooding to evacuate three hours before high tide.?

Thirty-mph gusts and beach erosion are the biggest concern in Sandwich, Mass., where residents boarded up their homes against the approaching storm.

?We had serious flooding this morning,? Brian Gallant, director of the town?s emergency management, told NBC affiliate WHDH on Thursday. ?We had a couple of roads that were under water. The boardwalk again is under water.?

Waves as high as 23-feet with 3-foot storm surges were expected Friday, WHDH reported.

Commuters slid into work on wet, sloshy snow in New York and New Jersey, with flurries expected to continue through noon before changing to rain, NBC New York reported.

A winter storm warning was in effect through noon in New York City and through parts of northeastern New Jersey, with 1 to 2 inches of accumulation expected in the city, the Weather Channel reported. A half a foot or or more could fall over northeast New Jersey and into the lower Hudson Valley, the National Weather Service reported.

Fifty-eight flights had been canceled for New York?s LaGuardia airport and 54 at Logan in Boston as of 9:30 a.m. Friday morning, according to Flight Aware.

The storm swept through the Midwest and up from the Mid-Atlantic earlier in the week, taking its toll in several states. A Virginia man died after his car slid off an icy road, and two North Carolina boaters remained missing offshore.

The other day I read this OUTSTANDING article on how to avoid affairs by a super couples and family psychologist, Dr. David Palmiter.? I have never seen an article like it! I love his candor and forthright advice on avoiding affairs.? Seriously, it is worth checking out.

Among his 10 tips, I especially liked the following:

Tip #2: ?Throw water on the spark. If you start feeling titillation towards another person do something to kill that. Putting some distance between you is always a good idea (e.g., stop having contact, make sure you are never alone together, don?t complain about your spouse to this person or encourage the same from him or her, avoid mixing contact with substance use).??

I love this! So simple to say, hard to do in some cases, but right on the money in terms of affair avoidance advice.? Sometimes the most simple actions are the more effective.

Tip #8: ?Reflect on what the pain from divorce is like.? Engaging an affair significantly increases the likelihood of a divorce and few human experiences are more stressful or painful than that.?

Divorce is unavoidable at times, avoidable at others ? but the subsequent pain is always there.? Particularly when children are involved.? Best to think twice, three times, then over and over again before starting an affair.

And my favorite, Tip #1: ?Be humble. Realize that an affair can happen to anybody.?

Nobody gets married thinking they will have affairs and get divorced, yet it happens everyday.? Not taking our partners and our relationships for granted is something for all of us to keep in mind.

Rates down as Spain sells $6.5 billion in debt

(AP) ? Spain has sold ?5 billion ($6.5 billion) worth of bonds at a lower cost, in another sign that investors are getting more hopeful over the government's management of the country's debts.

The Treasury said Thursday it sold ?2.44 billion in 10-year bonds at an average interest rate of 4.92 percent, compared with 5.2 percent in the last such auction Feb. 21.

It also sold ?2.01 billion in 5-year bonds at 3.57 percent, compared with 4.12 percent Feb. 7. It also placed ?569 million of 3-year bonds at 2.63 percent from 2.71 percent Jan. 17.

Spain's borrowing rates have fallen recently, partly on the back of the government's deficit-reduction program.

However, the country remains stuck in recession and has unemployment of 26 percent.

When it comes to discrimination, retaliation is still the No. 1 risk ...

Hopefully, most supervisors know it?s illegal to discriminate against employees and applicants because of their race, age, sex, religion or disability. But apparently fewer know that the same federal laws also make it illegal to retaliate against people for voicing complaints about such discrimination.

For the third year in a row, retaliation is the No. 1 type of job discrimination claim filed with the EEOC. Employees submitted 37,836 retaliation claims in 2012. That?s only a small increase from 2011, but up more than 100% since 1998.

Why the jump? The Supreme Court rewrote the definition of ?retaliation? a few years ago, making it easier to get retaliation lawsuits into court. And now many employees tack on retaliation claims to their age-, sex- or race-bias complaints to the EEOC.

EEOC rules clearly say that employers, ?may not fire, demote, harass or otherwise ?retaliate? against an individual for filing a charge of discrimination, participating in a discrimination proceeding or otherwise opposing discrimination.?

Advice: Don?t give employees any reason to file retaliation claims. Before taking disciplinary action against people who?ve complained about discrimination (either formally or informally), supervisors and HR should ask themselves these two questions:

  1. ?Why am I taking this action now?? Scrutinize the reason and timing of the action. Is there any connection to the person?s complaint? If it smells even a bit like retaliation, a jury could see it that way, too.
  2. ?Would I take this action with my best employee?? If your answer is ?no,? you could be open to a retaliation charge. If your answer is ?yes,? make sure that you document the basis for your decision before proceeding.
